miTarget™ 3′ UTR miRNA Target Clones 將miRNA目標基因的3' UTR基因序列插入在firefly luciferase (Fluc) 或Gaussia luciferase (GLuc) 螢光素酶下游處，由上游啟動子SV40驅動此段基因的轉錄。當miRNA辨認到3' UTR序列並抑制Fluc- 或 GLuc-3' UTR的轉譯，螢光素酶的表達受到抑制。藉由偵測螢光素酶的活性，可以得知目標基因是否有被miRNA調控。

miRNA target驗證、預測目標miRNA target site 的功能和miRNA對target gene調節作用的研究

LIN28為已知的miR-125a目標基因，
將GLuc-LIN28-3'-UTR clone轉染到
HEK293細胞，轉染(transfection)後
24小時，偵測GLuc和SEAP的活性。
將只轉染LIN28 3'-UTR target sequence expression clone的GLuc和SEAP的活性比設定為1，
作為標準化的基準。

 miArrest™ miRNA Inhibitors

當miRNA inhibitor construct被轉導或轉染進入細胞後，轉錄出的miRNA inhibitor會再經過轉錄後修飾(post-transcription)形成一個可以捕獲miRNA的結構，此結構可以和兩個成熟的單股miRNA進行特異性結合，阻止內生性miRNA與目標mRNA的結合，從而抑制miRNA對目標基因的調控，並且增強目標基因的表達。

應用
抑制miRNA以進行功能缺失(loss of function)的研究

將miArrest™ inhibitor clone轉導入細胞後可以阻斷miR-125a對目標基因的抑制作用。
使用劑量越高阻斷效果越好。在最高劑量下(100 ng)，GLuc活性已高於對照組，此現象
可歸因於vector-based inhibitor可能已經阻斷內生性miR-125a的調控作用，導致
GLuc-LIN28-3'-UTR transcript的轉譯活性增加。
