EZShuttle™  ORF Clones

——  EZShuttle™ Gateway® PLUS ORF Clones  ———————————————————————————

EZShuttle™ Gateway® PLUS ORF Clones的ORF序列兩側具attL1 / attL2位點,可使用EZShuttle™ recombination cloning system 或Gateway® systems進行Insert的轉移。兩側的multiple cloning sites (MCS),亦可允許使用限制酶切割連接方將Insert在不同表現載體上進行轉移。
 
  • 彈性的subcloning選擇 –可使用Gateway、EZshuttle系統或傳統的限制酶切割連接方式轉移Insert。

GLuc-ON™ Promoter Reporter Clones


GeneCopoeia GLuc-ON™ promoter clones使用分泌型的Gaussia Luciferase (GLuc)作為報告基因,可用於活細胞實驗、及時偵測promoters功能,並具超過20,000 個human或18,000個mouse已建構好的現成promoter clone或客製化promoter clone可供選擇。
 

Why use Gaussia Luciferase (GLuc)?

Gaussia luciferase (185 aa, 19.9 kDa)為目前發現最小的luciferase 1,可催化coelenterazine的氧化,此反應過程中會散發出冷光(480 nm),常被用在各式的Reporter Assay中當作 Reporter gene。
GLuc具以下特性:
  1. 為天然分泌型蛋白3,可直接從細胞培養基中收集,不需打破細胞。收取完後細胞可繼續培養,利於活細胞時間點實驗。
  2. GLuc所發出的冷光訊號可比Firefly 和 Renilla luciferases高一千倍,為實驗提供較高的靈敏度2
  3. GLuc可穩定存在於廣泛的pH值範圍及不同的細胞培養條件中2,4
