GeneCopoeia GLuc-ON™ promoter clones使用分泌型的Gaussia Luciferase (GLuc)作為報告基因,可用於活細胞實驗、及時偵測promoters功能,並具超過20,000 個human或18,000個mouse已建構好的現成promoter clone或客製化promoter clone可供選擇。
Why use Gaussia Luciferase (GLuc)?
Gaussia luciferase (185 aa, 19.9 kDa)為目前發現最小的luciferase
1,可催化coelenterazine的氧化,此反應過程中會散發出冷光(480 nm),常被用在各式的Reporter Assay中當作 Reporter gene。
GLuc具以下特性:
- 為天然分泌型蛋白3,可直接從細胞培養基中收集,不需打破細胞。收取完後細胞可繼續培養,利於活細胞時間點實驗。
- GLuc所發出的冷光訊號可比Firefly 和 Renilla luciferases高一千倍,為實驗提供較高的靈敏度2。
- GLuc可穩定存在於廣泛的pH值範圍及不同的細胞培養條件中2,4。
